چکیده

ETHALLY irradiated dogs usually recover promptly if given several billion fresh, autologous, marrow cells.1’2 Autologous marrow, stored at low temperature in glycerol or dimethyl-sulfoxide, induces a similar recovery.35 Recent studies of lethally irradiated mice have shown that infusions of fresh, isogenic leukocytes, if given in sufficient number, are followed by survival and return of marrow function.#{176}” Atkins, Goodman, Perry and Kerby10 2 have shown that glycerol exerts a protective effect on blood leukocytes stored at low temperatures. This communication reports the survival of dogs given x-irradiation in a dosage of 1200 r and infusions of autologous leukocytes that had been stored at -80 C. in 10 per cent dimethyl-sulfoxide.#{176}

Recovery of lethally irradiated dogs following infusion of autologous marrow stored at low temperature in dimethylsulphoxide.

By JOHN A. CAVINS, SHINPEI KASAKURA, E. DONNALL THOMAS AND JOSEPH W. FERREBEE D OGS lethally exposed to general radiation usually make uneventful recoveries if given prompt infusions of a few billion autologous marrow cells.1’2 Cells for these infusions can be preserved indefinitely at bow temperature in glycerol. ’4 Recent studies suggest that dimethyl-sulphoxide may be preferable to glycerol ...

Regeneration of blood-forming organs after autologous leukocyte transfusion in lethally irradiated dogs. II. Distribution and cellularity of the marrow in irradiated and transfused animals.

Dogs were given transfusions of cryopreserved autologous mononuclear blood leukocytes after 1200 roentgens (R) (midline dose) whole-body x-irradiation. Bone marrow repopulation was studied by means of histomorphological methods at days 9 and 10 after transfusion of an average of 3 X 10(9), 7 X 10(9), 13 X 10(9), and 31 X 10(9) cells. The return of marrow cellularity to normal values was related...

Engraftment of dogs with Ia-positive marrow cells isolated by avidin-biotin immunoadsorption.

Previous work has shown failure of engraftment in lethally irradiated dogs when autologous marrow was depleted of Ia-positive cells with an anti-Ia antibody and complement before infusion. In the current study, we have utilized an avidin-biotin immunoadsorption procedure to obtain a population of highly enriched Ia-positive cells for autologous bone marrow transplantation in dogs given lethal i...

Novikova MN, Nemenova NM, Malanina VN: Hematopoiesis in long-term survival following supralethal irradiation and bone marrow transplantation in dogs.

B ONE MARROW TRANSPLANTATION may effectively protect dogs exposed to whole-body ionizing radiations of 2 to 3 LD95. Hematopoiesis was investigated at remote periods after the exposure, when the hematopoietic stem cells had been proliferating in the lethally irradiated organism for a long time. Ferrebee and his associates’6 succeeded in attaining long-term survival of supralethally irradiated do...

Prevention of transfusion-induced graft-versus-host disease in dogs by ultraviolet irradiation.

Ten dogs were given 9.2 Gy of total body irradiation and autologous bone marrow infusion followed by ten daily transfusions of leukocytes for a total of 11.5 to 36.2 (median, 18.8) x 10(8)/kg obtained via leukapheresis from histoincompatible unrelated donors. Four dogs were given unirradiated leukocytes, and all developed graft-versus-host disease (GVHD). In contrast, only two of three dogs giv...
